May a commissioners court regulate game rooms within the county?

Yes. A county may restrict game room locations to specified areas of the county, prohibit locations within a distance prescribed by the commissioners court to a school, place of worship, or residential neighborhood, or restrict the number of game rooms that may operate within a specified area of the county. Local Government Code §234.133. See this question and more in our Bingo, Raffles, Gaming Rooms and other Gambling Issues under Texas Law mini-publication.
